I’m planning to co-culture 4 different cancer cell lines and label each cell line with different dye to differentiate them, then perform a drug cellular uptake experiment. Is there a device I can utilize to quantify the amount of drug uptake by each cell line?...protocol?
It would be easier if you make four different cultures.
Then add the drug in the same concentration in each culture.
Finally you measure drug concentration in the four different cultures media. Supposedly the difference among cultures will show you roughly the difference in uptaking.

You can culture different cell lines separately. You can incubate each cell culture with a known amount (example 1 uM) of a drug dose for a know time (example : 30 minutes). Then change the media and analyze the media spectrosopically. You may definitely need to do replicates in this experiment. Many drug molecules have optical absorbance (or even emission sometimes). I would suggest you tag the cells with different fluorescent proteins such as YFP or RFP for the differentiation of cells from each other before the co-culture and than use different fluorescent dye to tag your compound of interest to estimate the uptake of ur drug, such as FITC and analyze through confocal and flow cytometry.
