Generally, in the G phase of cell cycle cell growth occurs. Due to S phase, replication of DNA as well as synthesis of proteins require for cell division to occur. So, is it possible to increase the cytoplasmic content of the cell without any further cell division?

In some cell types, such as keratinocytes (main cell type in epidermis), just blocking the G2/M transition with nocodazole (or bleomycin) triggers a series of molecular events that culminate in endoreplication : cells continue replicating their DNA, but they don't divide. One consequence is cell growth, along with polyploidy of course.
This might be limited to cells having a propensity to endoreplicate though (keratinocytes do), since doing this in fibroblasts just results in apoptosis (or at least cell death, I don't remember exactly).

Interesting question. What cell type are you working with and what do you want to achieve? As mentioned earleir, it is not so difficult to prevent cell division. If ployploidy is no problem, I can give you a number of treatments to prevent cell division. If you want to prevent DNA synthesis, no problem, I have several protocols for that as well.
Thank you for the opinions. I am working on mESCs (mouse embryonic stem cells). I have observed that when mESCs maintain the pluripotent state, the cell size remain small compared to the differentiated state, where the cell size is more. So, in that context cell size is cell specific. Basically, cell size matters and plays a significant role to promotes the cell towards further cell cycle phase and division. In tumor cell, same sort of phenomenon also can be seen. Uncontrolled growth as well as proliferation is observed in tumor cells. So, If I want to grow the cells in a extended level without cell division and polyploidy, how should I achieve it.

I think to get a really helpful answer you need to tell us what you want to achieve. Do you want bigger cells, more protein, more DNA content, etc..? And to what end? As some people stated above there are different methods/chemicals that will cause the cell to behave in an abnormal way (i.e. e.g. multiplying DNA without dividing), but to find out what's best for your experiment you/we have to know what exactly you want to achieve or what you mean by "to grow the cells in a extended level".

try mitomicin treatment, it will inhibit cell division but does not affect cell growth. Let me know how this goes:)
Retinoic acid promotes differentiation in some cells. Also elevating cAMP, so forskoline treatment maybe?
