I used 26 gauge needle for subcutaneous injection of matrigel:cancer cells. However, when i pull out the needle, the matrigel solution also came out. How to prevent the leaking?
Your sample may not be gelled enough or too close to the hole. Inject the mice and hold the syringe in place for at least 15 s to allow the gelling to start. Use a longer needle for injection and inject all the way to the end of the needle so that the needle tip is as far as possible from the exit hole. And rotate the syringe as you withdraw the needle. This will seal the hole and reduce leaking of liquid, but there is still likely to be some leaking.
1 ml syringes with 22 gauge needles can be substituted . try slowly pull up 100 μl of cells alone or 200 μl of cell/matrigel mixture and make sure to hold the needle inside for a few minutes. Also the mouse's body temp allow the matrigen with cells to solidify quite quickly.
There is a tip I learnt when I was doing such work: lift the mouse skin up after pinching and carefully move the needle right-left to create space and then deliver the suspension while making sure that the needle still holds the mouse skin up and the space you created is not deformed.
