I would like to make a complex of manganese with edta in water. Until now i just tried 0.5 mM of the acid EDTA and 0.5mM of MnCl2.4H2O in 100 mL solvent water, put the reaction 37°C for 3 hours pH=5,5 but no color was formed and no absorbance was observed in the spectrum.
I am looking for a manganese-EDTA complex to quantify at 400-500 nm of absorbance.
Anyone have a relieble protocol with MnCl2 to make manganese-EDTA complex?

It's been a few years since I worked with any chelates, but I recall pH is a massive influence on the reaction. If you google "edta metal complex pH" some of the hits include lecture notes on complexation chemistry eg:

So you might need to adjust the pH to ensure complexation is occurring.

as Prof Matthew mentioned some factors such as pH are very important , I think according to the reference which is attached , the proper pH 10.5 with controlling other factors , you can manage it easily!
