I am working with very limited tissue/cell samples and need to analyze both mRNA (RT-qPCR) and protein (Western blot) from the same sample.

  • Is there an efficient protocol that allows sequential extraction of RNA and protein from the same lysate (e.g., TRIzol or other combined methods)?
  • Would you recommend splitting the sample in advance, or is it better to process everything together and then separate phases?
  • Are there any kits or optimized workflows that minimize sample loss?

I would appreciate advice from those who have successfully done this, especially when the sample amount is small.
