I have a fluorescent sample and want to measure the excitation and emission wavelength using the Synergy HT BioTek microplate reader. I have tried spectrophotometer ND1000 and 3300, but want to figure out a way to get the same result from the microplate reader.
Does anyone know how to do that?
I have defined a protocol for reading at 360/40,460/40; 485/20,528/20; 540/25,590/35; 635/32,680/30, have got some results using the defined protocol in Gen5, but don't know how to interpret them.
Any help is highly appreciated.

Dear Nelly, In a micro plate the fluorescence emission optics are very different from those in a single cell because of obstruction, reabsorption etc.
Therefore you need to ask a physicist if some configuration, e.g. of full/empty cells, and illumination might make this possible, Paul.
