I want to determine the conjugated drug to IgG ratio by nanodrop. I know the extinction coefficient of IgG and drug by M-1cm-1 but all the unit in the nandrop program is based on g/ml and so on (attached fig). How can choose this unit?!
Thank you Dr John Carter for your reply. Actually, I want to input the Extinction coefficient number and it's critical for this measurement. The unit in user-defined also is similar in the figure.
OK. So you just need to convert the M-1cm-1 to g/mL. If your Ab is an IgG you may safely assume the MW is pretty close to 150,000 g/mole. (And realistically, you're not going to get much better than two significant figures in your assay.) You're probably using a standard 1 cm long/thick cell in the spec, right? So that's =1 in your calculation.
