I need to use an anti fungal and have got cycloheximide, is this right?
0.5 mg/mL stock solution - autoclave 200 mL water then add 100 mg Cycloheximide powder and mix
After autoclaving agar, add 10 mg/L of stock solution, so 500 ml agar, add 10 mL of 0.5 mg/mL stock solution
Is that concentration enough to eliminate most environmental fungi or do I need it stronger?

your final cycloheximide concentration is pretty high at 10 mg/L. I found some data on cycloheximide toxicity regarding a few fungi here: http://antibiotics.toku-e.com/antimicrobial_548.html
I think that the concentration you use is sufficient to inhibit growth of most fungi.

Thank you, I've read some usually use 20 mg/L, which is obviously stronger than mine, is there any negative effects if I use to strong concentration?

It depends on your experiment whether 20 mg/ml cycloheximide might be too high due to some unwanted side-effects. You could do some preliminary experiments with the lower conc. of 10 mg/ml. If this should prove insufficient increase the conc. to 20 mg/ml. Hope this helps you.
If your final cycloheximide concentration is 10mg/L (which is equal to 10µg/ml), I think that's low to eliminate fungi. I've seen many papers discussing on cycloheximide resistance in fungi. Some of them can tolerate to 500µg/ml of cycloheximide. In our lab, we always supplement our culture media with 100µg/ml cycloheximide and it works in killing fungi and eukaryotic algae.
@Kier Klepzig: Safety data sheets for cycloheximide usually recommend to dispose of it in accordance with local, state, and federal regulations. The best way would be to check back with the person responsible for waste disposal at your institute.

yep. Saw that already. No such regulations to be found for these specific compounds. We will neutralize through autoclaving.
