I am working on leukemia stem cells now and trying to get stem cells from leukemia cell lines, like Sup-B15. I've used FACS targeting CD34+ and side population assay, however no positive cells were detected in both cases. Since Sup-B15 grows in suspension and forms sphere, it's not likely that I can use colony formation assay (Or am I wrong? ) It's generally thought that CSC are much easier being extracted from leukemia than solid tumors. Is there any other way to isolate stem cells from this cell line?  Thanks a lot!
