I am performing the mitostress seahorse assay in aortic endothelial cells. I am looking for a good positive control that would increase the glycolysis/glycolytic capacity when I treat the cells in vitro to compare it with my treatments.
Anyone who has ideas about it, your help is appreciated. Thanks!

the composition of the seahorse medium for mitostress or for glycolysis is slightly different in the final amount of glucose, glutamine, and pyruvate. Therefore, I would suggest not looking at ECAR or OCR in the same experiment. In my experience, I used insulin (100nM) in C2C12 cells (muscle cells) to induce glycolysis and to test if it was still boosted with oligomycin. Check out this paper it can be useful.
