At first, I am planning to de-methylate (existing methylation) my culture with 5-azaycytidine drug and want to inactivate the further action of this drug. Then I am planning to infect my culture with respective organism to study how far DNA Methylation has induced due to infection alone particularly.
Finally, I want to extract DNA, RNA for Methylation and gene expression studies.
i.e. I want to study the effect of DNA methylation only because of infection in my culture. This is my plan actually.
So, experts plz do correct me if I am wrong, the way I planned above also aware me how can I carry out my experiment with the protocol as I mentioned above.

You can use Epi info program and calculate sample size according to type of study and prevelance of disease
