I agree with Jordan: monocyte morphology change dramatically after macrophage differentiation, the former looks like a round cell and the latter tend to spred with cytoplasmic looking pseudopods. A function test without doubt is phagocytocis of latex beads, red bllod cells, or CANDIDA and Staphiloccoci bacterial cells. You may also use flow cytometry using fluorochrome label monoclonal antibodies against macrophage CD markers. Good luck.
