Recently, I am working on a project about leukemia. Here, could anyone kindly tell me how to establish a human leukemia model by employing K562 or Jurkat or other human leukemia cells in mice? Any suggestions or references are welcome!

If I understand your question correctly, you would like to grow one or more of those leukemia cell lines in mice. In other words, you would like to establish a cell line based leukemia xenograft model. For this purpose, you can inject intravenously 5-10 million cells (if the cells have never been passaged before) into sublethally irradiated SCID mice or unirradiated NOD/SCID mice (while there are alternatives such as the use of NOD/SCID gamma mice or intratibial injections, they are a little more difficult and expensive). You could use RAJI (Burkitt's leukemia), NALM-6 (pre-B leukemia), RS4;11 (pro-B cell leukemia), MOLT-3 (T-cell leiukemia), just as a few examples. Within 90 days mice will develop morbidity and when you terminate them you will find enlarged spleens and leukemic cell infiltration in multiple organs, especially liver, bone marrow, kidney, and CNS. You can then take the spleen cells and use them in your future experiments. You would need fewer cells to inject with such cells. Spleen size should be 3 com or more before you decide using such cells. But it might be better to use primary leukemia cells or transgenic mouse models depending on the questions you are trying to answer in your project. U can also do a sub cutaneous injection with matrigel in NOD/SCID or SCID without irradiation but requires 1 million to 10 million cells and it it develop into a tumor in the SC area.. but it somehow migrates to spleen and BM (only organs i have looked at)...
I.V injections are better, Im have not done with k562 but i have done with NB4 in scid without any sublethal irradiations, at the rate of 50000 cells per mice, it took 40 - 50 days for the SCID to die with enlarged spleen.
