I purchased 200kDa Hyaluronic acid (HAc) powder and want to make a hydrogel for dermatological fillers with it.

In many kinds of literature, it was confirmed that the gelation process was performed using a crosslinking agent called BDDE for the gelation of HAc in NaOH.

With this in mind, the experiment was conducted as follows.

1. Completely dissolve 10% HAc in 0.25N NaOH. (120rpm, 30 minutes to 1 hour)

2. Add 1% BDDE crosslinker to the completely dissolved HAc solution and continue the reaction. (40 ºC, 120 rpm (magnetic bar), over 6 hours)

It is said in the literature that gelation is easily achieved through this process.

But, I do not have gelation.

Is there something wrong with my method?

*As you can see in an uploaded picture, it is in solution even after 24 hours, and it only changes from transparent color to brown, but gelation does not proceed....

* Is it correct to continue mixing with a magnetic bar during the crosslinking process?
