I prepared silver nano particle using AgNO3 with the plant extract.they are in poly dispersed phase. it is difficult to separate.whch is the best solvent for antimicrobial ativity?
One year ago I conducted few experiments concerning determination of AgNPs activity against bacteria and bacterial biofilms.
MIC was determined using broth microdilution method on 96 well plates in MH Broth according to CLSI (Clinical and Laboratory Standards Intitute)
MBEC (Minimal Biofilm Eradication Concentration) was determined also in 96 well plates. Visualisation of the results was carried using resazurin as cell vaibility indicator.
Compounds In appropriate size (nm) where dissolved in PBS.
I agree with Maciej Jaskiewicz, and I suggest you to take a look at "Bacterial effects and protein corona evaluations: crucial ignored factors in the prediction of bio-efficacy of various forms of silver nanoparticles".
thank you for your suggestions.actually here plant extract reduce the silver and form a complex with Ag .There are lot of publications in it. I want do know how to dissolve the compound
I suggest first you calculate the concentration for your silver nanoparticles then well diffusion method or disc diffusion method then you can determine the MIC.... Attaching some paper hope it will help you ..... or you can download from my publication list..... all the best
just go for antibacterial activity. make a petriplate with agar. make some holes and pour NPs solution. clear zones will show antibacterial activity of AgNPs.
Choose species in gram positive and gram negative .Prepare medium of your comfort and incubate it. Analyse using disc diffusion as it is simple. Keep measure of Ag Np gram used for disc.Calculate disc size and cross check with corresponding gram.
I think should do . Get more suggestions and try them too.
