I want to use Propofol, highly hydrophobic, in my cell culture media for human brain microvascular endothelial cells (HBMEC).
My stock propofol is 10 mg/ml (Mw: 178.271 g/mol)
The desired final concentration of propofol in my media that I need for exposing cells with, is 50 uM.
I'm wondering how I could dissolve propofol in DMSO and then in my media, by considering that the final concentration of DMSO should be 0.5% to avoid cytotoxicity.
I'd really appreciate it if you could help me with this!

The molecular weight of propofol is 178.271 g/mol.
So 10mg/ml stock solution will be 56.17mM.
If you need to make the final working concentration of 50uM in (say) 2ml of cell suspension from the stock of 10mg/ml (56.17mM), then you will have to add 1.78ul of stock (10mg/ml) which is dissolved in 100% DMSO (make sure the compound is dissolved completely in DMSO) to 1998.22ul of culture media. This will correspond to less than 0.1% DMSO (0.089% DMSO) in the final cell suspension which is non-toxic to the cells.
If you are to use less than 2ml of cell suspension in the experiment, for instance 96-well format then you will have to dilute the stock (10mg/ml) with culture media accordingly, say (1:10) in order to avoid errors in pipetting small volumes of stock solution.
So, in either case you will have DMSO concentration less than 0.5% in the final culture media that is being exposed to the cells.
