I am working on a magnesium based alloy, and i wanted to evaluate the cytotoxicity of my alloy with MTT assay. I want to know the reason why most of the researchers perform the MTT assay in three time points, like 1, 3, 5 days. In ISO 10993-5, examination of the cells to determine the cytotoxicity is after 24h to 72h.

First, I want to know if it is necessary to perform the test in 24h AND 48h AND 72h? Or it is possible to perform it just for one time point or two?

Second, is there another reference which contains the time points of 1, 3 , 5 days or 2,4,7 days (these are the time points I saw in papers)?
