I have to stop my enzyme reaction pH(8.5) by perchloric acid and further neutralized by KHCO3. How to determine the amount of acid that is required to stop the reaction and is there any calculation for this .Because it is a coupled assay, first I have to stop and neutralize it and proceed for the second reaction.

In one of the papers it was mentioned that 3% of the perchloric acid was used to stop the reaction pH(7.2) before neutralization with KHCO3.

Please give you suggestions..
