First you should download est from the database then use vec screen to removr vec sequences and polyA tail make contings using cap3. If you are well versed with pirl script use MISA TOOL to locate ssrs in the unigenes using primer 3 or batch primer
I don't know in which stage you are of processing the EST database. As Padder suggested, if you have the raw data first you need to do the quality control and then assemble the sequences in order to obtain EST contigs. For the assembly there are many programs, but I would suggest Mira or Newbler. Once you have the contigs, you can try to use msatcommander (http://code.google.com/p/msatcommander/) to search for microsatellites. This programs lets you choose the kind of repeats you want to find and also it uses primer3 to design primers in the ssr flanking regions.
Specifically for differentiating between male and female plants, I can't help.
If you have the EST contigs as Sara mentioned you can also locate quality SSRs using sputnik (http://espressosoftware.com/pages/sputnik.jsp) and then design your primers flanking the SSRs using FastPCR (Kalendar et al., 2009)
