I just purchased one vial of Raji B cells from ATCC. It is a suspension cell. Should I culture it in non treated culture flask? or can i use normal tissue treated flask? And how to assess the cells morphology under microscope?
I would just use regular TC plates. They will not stick regardless since they are suspension cells. To look how they are doing put them under the scope and gently shake the flask, you will see how they are floating and what their morphology is like from that. They should look nice and round. when they are confluent, the media will be completely cloudy when you look at it from the bottom.
For Raji, RPMI medium 1640 + 10% FBS + 100u/ml Penicillin + 100 micro-g/ml Streptomycin + 2mM L-glutamine is fine. Remember to change the medium every 2-3 days.
