Recently, we have done the macrophage antigen lpresenting experiment by co-culture of macrophages pre-loaded with OVA and CD4+/CD8+ T cells isolated from OT-II/OT-I mouse spleen. Using CFSE to stain the T cells, after 3-5 day co-culture, there is no significant differentiation of the target T cell.
Is there any probelm in the experiment? and is thre any trick in processing the sample?

You mean that only CD25+ T cells isolated from OT mice immunized with OVA injection i.v. can be proliferated when co-cultured with macrophages primed with OVA? the remaining CD4+ or CD8+ T cell can not be proliferated? Another questions, the macrophages we used should derived from the same OT mice immunized with OVA injection i.v. or any other naive mice or any other mice immunized with OVA injection i.v. ？Thank you very much.
