It is my first question here, so I hope you can help me..
For an experiment of my thesis I provided pure cultures with different concentrations of lactate an monitored them over time using HPLC. I now have the Lactate concentration within the medium for the pure cultures, as well as for abiotic control bottles. Both show a decline in lactate (stronger in inoculated bottles), but how can I know calculate the lactate consumption rate of the bacteria? What is the best way to display bacterial lactate consumption? So far I never did such plots and am very confused what to do next.

Just measure it spectrophotometrically using a kit or automated blood lactate analysers or strips .....Many methods are available, you can also measure a decrease in lactate concentration via lactate metabolizing enzyme.

The substrate consumption rate will depend on the initial concentration of the substrate, S0. If you subtract the residual substrate at each time point from the preceding one, you will have the amount of substrate consumed from one time point to another (Sf-Si=dS). If you divide dS by the time interval dt, then you will have the consumption rate at each time interval. Note that consumption of lactate will ensue in biomass increase. That is, the bacterium is supposed to have some of growth on lactate. Therefore, you could do a growth curve showing changes in biomass against time. You could do a pliot showing both substarte consumption and biomass accumulation on one graph
