The screening of this particular protein molecule is only from normal samples (not entail any pathological samples). How to calculate the sample size in this context as this study does not involve any situation such as standard deviation, comparison of means for significance, etc. Thanks in advance for the experts' input.

Sir, thank you for the reply. Our research question is to decide whether the protein of interest is expressed or not expressed on the human spermatozoa (only from normal subjects, won't include the samples from diseased subjects). The semen samples will be collected from the volunteers, and screened for the routine sample analysis (normal count, motility, vitality, and morphology). Only the normal samples will be considered, other samples will be dropped out. Then, the normal semen samples will be processed to get the washed sperm pellets, used for the further downstream experiments such as Digital PCR (to measure the transcripts of the gene of interest of protein), and Immunoblotting, & Immunocytochemistry (to confirm the presence/absence of the protein of interest).
