I'm doing a projekt where I have to try to mitigate an interference from anti-drug antibodies when measuring my drug.
My plan is to use AlphaLISA og another kind og ELISA method
Apart from acid dissociation does anyone have some idea of how a kan do this? an alternativ way? also any tips on how to do the acid dissociation is very welcome
If your drug is not temperature sensitive then technically the antibodies can be denatured by heat. Simply keep the samples at 80 degree centrigrate for 1h and your should be free of bound antibodies.
Alternatively, you may also do protease digestion of your samples. I believe proteinase K should digest antibodies.
@Suhad, Thanks but that artikel only talks about how to prevent ADA forming in the body not how to mitigate the interference from anti-drug antibodies present in the blodsamples.
I'm trying to mitigate the interference so that I can measure the "real" amount of my drug in my samples

Unfortunately my drug (insulin) cannot with stand so high temperatures
