Hello Everyone

I was trying to measure a protein in human urine samples using ELISA. When I was doing spike-recovery experiment, the recovery is less than 50 %, indicating that urine matrix is interfering.

Can we avoid the interference by diluting the urine samples ? If yes, what should be the diluent used ? what should be the factor of dilution ?

Also, what should be the blocking agent used to reduce interference ? I am using 1% BSA. Is milk better than BSA ?

Is there any other way to avoid interference ?
