One of my student needs to check the chitinase activity of Trichoderma spp. But she is not able to make the medium containing chitin. She is using chitin from shrimp shell (Hi Media). Even after adjusting pH the media remains liquid. Can any body plase help me with cellulytic and chitinas eactivity protocol?

As my experience, liquid media are the best ones to produce extracellular enzymes of fungi. If it was appropriate, you can extract the enzyme in both media and mycelium.

If you want to try to find colonies that are able to conduct chitinolytic activity you can check out my old paper, link below. We did not have trouble as we used a semi-solid medium for growth. Regions of breakdown (diameter of clearing) could give you a relative indicator of chitinolytic activity, and then for more detailed studies a liquid (or semi) could provide greater results.
