I am currently looking at the best method to quantify osteoblasts. According to the ASBMR guidelines (see below), we can quantify them by OsteoMetrics system, which is basically counting the cells that are residing on the bone surface as osteoblasts through H&E.

This method is widely accepted and being published in many high impact journals, however it doesn't seem to differentiate between active and inactive osteoblasts (or mature vs immature). We tried IHC using osteocalcin antibody but we are not quite satisfied with the results (strong background staining, possibly due to osteocalcin being secreted). Is there anyone with expertise that can provide advice on this? Thanks!
