I understand a glycerol stock for long-term preservation would be ideal, but how are working cultures generally stored? Mainly concerning Candida spp. isolates. A few references mention that they can be stored in SDA at room temperature, but how long?
Hello, mainly storage in clinical laboratories in glycerol stocks, but for interesting fungi you can grow in reach media, than you can put in freezer for sporulation after you can collect spores and keep in dried conditions as long as you want.

You can store them on SDA or other suitable media slants in test tubes in the fridge.
We used to use screw-capped tubes, inoculate them with working cultures, and store them for a month in a fridge.
If those are not available, simple microbiological caps are fine too as long as the media is not drying out. We never used cotton-plugged tubes since molds do overgrown it.

the candida spp genomes are highly plastic and unstable even at 4C, it is therefore not ideal to stock the strains on plates for too long, to avoid accumulation of mutations. usually not exceeding 1 week.
