I want to synthesize cDNA from plant RNA. Please tell me that what should the cDNA band look like on gel, so that I can guess that cDNA has successfully synthesized. Please show me some gel pictures for this.
In general, it's not necessary to run an agarose gel for this. And if you do, both RNA and DNA will be visualised. Instead, you should run a qPCR assay with your newly synthesised cDNA and -RT (no reverse transcription control, that is a reaction with the same composition as in your cDNA synthesis reaction minus the Reverse Transcriptase) samples. Ideally, you will have amplification in your cDNA sample and no amplification in your -RT control. However, if you have amplification in your -RT control, this may indicate contamination of your RNA with genomic DNA or contamination of reagents.
