U R WORKING WITH PLANT or ANIMAL TISSUE ? , i suggest if u use fresh material of (plants or animal tissue) like 1 -2 days old stored material at -20 c. but liquid nitrogen helps prolonged storage. Use lqd nitrogen , i guess u r not tired of using lqd ntgn for crushing ur samples ? lqd ntrgn stored samples are good to be used for TRIZOL METHOD. if u r using freshly extracted samples witthout lqd ntrgn then no problem in RNA isolation will occur with trizol.
Few people that I know extract RNA with kits but the use of Liquid N goes without saying. How will you crush the sample? It is needed to transfer the crushed sample into a vial for further processing, when using a kit.
Yes we extract very good quality RNA from various species of Brassicaceae plants using Macherey Nagel, Fermentas and Qiagen kits without the use of liquid Nitrogen. We homogenize the tissue using either micropestles or tissue lyser. But for certain woody tissues like roots, liquid nitrogen works the best. Best of luck.
I tried RNA extraction using commercial kits several times without using liquid nitrogen. Qi-gene is a my preferable company to provide suitable RNA extraction kit specially for cancer cells. Always I used their product and got good result. Please kindly try kid. TQ

I used this kit to extract virus-RNA from a single aphid. I worked very well. My downstream application was RT-PCR.
@Nadine you are right but I guess us Pakistanis are used to getting things the hard way because of scarce resources and funds so for us Trizol-X all the way lol
