How should I induce phosphorylation of over-expressed wild-type alpha-synuclein (and its mutants) and successfully detect them in vitro?

After the over-expression of wt alpha-synuclein in vitro, I am unable to detect phosphorylated (S129) form of alpha-synuclein (antibody used - EP1536Y). Although I can see very few inclusions, but they are not positive for phospho aSyn. What should am I missing, and what should I do?
