I had mammalian cancer cells cultuted and fixed on confocal slides. I could see these small small bodies stained with dapi. The main culture flask doesnt show any bacterial growth. Is this mycoplasma? If yes how do I get rid of it?
Mycoplasma are not visible under a standard tissue culture microscope so you won't be able to spot them in the main culture flask. Are you testing for mycoplasma somehow? For example by PCR?
If you do have mycoplasma contamination it is difficult to clean a culture and if possible best to trash it and start from a new stock.
Hi, to be honest, it is really difficult to tell, especially as it seems there is a high background noise in general in the images.
It could be some kind of contamination, it could also be that the cells weren't properly washed after staining. Furthermore, depending on how you permeabilize the cell membrane, mitochondrial DNA also becomes accessible for the DAPI stain.
