I want to immobilize enzyme on chitosan macrobead, i want to know macro size of particle doesn't have any advantages for immobilization? I use CMC as substrate,can I relate this viscose substrate has limitation access to enzyme in micro or nano size bead?
It seems tome that you would choose the size of the particle based on its intended use. If injected into a mammal, nanoscale (~100 nm) particles accumulate in tumors and sites of infection, whereas microscale particles accumulate in macrophages and the liver.
Ok Adam,but my protein is industrial enzyme.for accessing to viscose substrate,can macrobead operate better than micro or nano size in activity?it is only my speculation!do you have any idea about using macro particle instead other?!!!!
This is not an area of expertise for me, but I have 2 thoughts for you. (1) The smaller the bead size, the greater the surface area-to-volume ratio, so the same volume of beads will have more immobilized enzyme if the beads are smaller. (2) Smaller beads will settle out more slowly than larger beads, so less effort will be needed to keep smaller beads in suspension.
thanks Adam for your idea,but exactly i want sth vice versa!!i imobilized enzyme on macrobead and i want to explain why i choose this size of particle?!you think macro size doesn't have any advantages to micro size?can i say large molecule like substrate can't access to enzyme molecules in pores and inside area of micro or nano size,so i choose macrobead!!!?!
Are the internal pores larger in macrobeads than microbeads, or are there just more of them?
I don't prepare SEM from bead,but as literature i can say in chitosan nanobead porosity is more
Mr Adam,can you guide me about porous particle!?can I say chitosan bead is porous particle?!I think, I immobilize enzyme on particle surface!so macro size can't be a good choice as surface to valume ratio is lesser than micro size!
I would guess that the porosity of the particles depends on the procedure used to make them. If they are not porous, then external surface area alone determines the amount of enzyme that can be immobilized on them. I see from the literature, however, that there are ways of making them porous.
