Does anyone know that how many cells lines and primary culture can be grown in L-15 medium? I need the complete list of cell lines and tissue types.
Our CO2 incubator in cell culture facility is not working and we need to grow our cells in L-15 medium which does not require CO2 for buffering. Another medium is CO2 independent medium by Thermofisher but the information about this medium is quite scarce.
medium using changes, so there is not likely to be a comprehensive list. L-15 buffers optimally at pH7.4 and a bit higher as it was developed for embryonic chick tissue, not mammalian cells. L-15 buffering uses extra amino acid content in addition to phosphate buffers. it works well for amphibian tissues, also. If doesn’t work for your cells, try CO2 the old-fashioned way: get a dessicator with an insert . Put some sterile water below the insert and bubble a little CO2 through tubing with a sterile , plugged pipette. Incubate your cultures within the dessicator in your incubator. CO2 will be released from the water and acidify your medium.
I know only one cell line MDA-MB-231 as recommended to grow in L-15 media. But they grow very well in DMEM also.
I would suggest using 10mM to 25mM HEPES to maintain pH in your standard media. 100x solution of HEPES is available from many vendors.
