As I conduct the Annexin V & 7-AAD staining experiment for cancer cell lines such as PC3, but I cannot decide the time when the positive control namely 5-fu would work? And could anyone know how to avoid cell death in negative control after treated with trypsin to detach adherent cells?

As a negative control, you can treat your cells with some apoptosis inhibitor (e.g. Z-VAD-fmk). For detail time-dependence information it is best to perform some time-lapse analysis. You can use various microscopic methods or some western blot or flow cytometry approaches in multiple time points.
