How long RBC can remain intact in whole blood collected in EDTA Tubes before being processed for Isolation of RBCs? I want to know conditions for storage of such samples so that they can be processed the following day. For how long can they be stored and under what condition?
In my center, usually we will isolate the RBC within 6 hours after we obtain from the patient and the sample will be kept in ice immediately after obtaining from patient till we start to isolate the RBC.
Dear it will best within 3 hours for the best integrity of the RBCs.
I recommend storing the RBC in the cold room for no more than 4 hours. But this recommendation is general; the storage time depends on what properties of the cells you plan to examine.
I do agree with Gregory comments.
EDTA- induced shape changes in cellular components of blood are pretty quick and reaching to plateau region in 2-3 hours.
In fact we did used successfully both cold and EDTA-induced shape changes to measure platelrt concentrate fuctionality, using automated cell counters, looking at changes in their cellular indices, based on paired sampling principles [ adding samples in empty container and dry EDTA containg tubes } to define the plateau of changes. In practice cold induced changes up to 8 hours are reversible. However EDTA_induced changes reached to plateau about 1-2 hours dependinf how fresh the products are and this function is substantially reduced after 24 and abolished after 3 days. While these data maily relate to PLT but by extrapolation might be useful to other cells and shorter storage time will be better if the shape changes are concerned

in short if you have modern cellular counter , particular based on light scattering principles, then looks for changes in cellular indices over the time and set up your own standard for different types af cell counters.

For my set of experiments, I prefer to separate the RBCs within 4 hours after collection in EDTA tubes. If you can store it at 4 degree C, the whole blood can be used for 2-3 days, though I have noticed morphological deformations of the erythrocytes even after storing it at 4 degree C. About 20% of this erythrocytes deform themselves to somatocytes (mostly) in a day or two.
Please also contact Dr jeffery Putter, as he is currently looking the issues of complex shape changes , deformabilty and micro vesicle formation of EDTA samples in both dark and light using a specially designed microscopic system. He is a member of reaserch gate and published some of his early findings

It is better to store bllod in 4 degree celcius and isolate RBC membrane immediately within 4 - 6 hours.

I think that you can lengthen the storage time of RBCs by the following procedure. At the first stage, isolate the RBC by centrifugation, at the second, prepare a cells-free (leukocytes and platelets depleted) plasma in which to store your RBCs.
Storage of blood at 4 degree is recommended. If stored in EDTA tubes, they retain their morphological shape upto 3- 4 days approximately.
