If your conditions are polarizing to begin with (i.e. you differentiate and polarize at the same time) then 6-7 days are plenty. If you first differentiate then polarize then it's 2-3 days (usually).
If tissue was fixed in 4% PFA then cryoprotected in 30% sucrose, are you able to wash and switch to FFPE processing instead of blocking frozen in OCT?
