I want to subculture a 75 cm^2 flask of MCF7 cells, grown in standard conditions (37 degrees C, 5% CO2), into a subdivision ratio of 1:4. How long does it take to culture these cells before they can be subdivided? Do I need to split the process into two subdivisions (i.e. divide one flask into two, and then two flasks into four), or can I subdivide all at once?

MCF-7 cell lines will reach confluent within 6-7 days in a 60mm culture dish. Then after reaching confluency you can do the normal cell passaging with sufficient amount of seeding density.
You can subdivide all at once.Once the flask reaches 80% confluency trypsinise the cells and seed them into other flasks.
It depends on how many cells are initially seeded. 75 cm^2 flask reaches confluency with approx 8-10 million cells. You can seed the cells accordingly.
