I need to perform an antibiotic selection after transfecting my cells, my questions is: How long can a transient transfection last for me to effectively select the successfully transfected cells.
In theory, it should be between 1 and 7 days, but in my experience it can be longer, at least 10 days, depending on the vector. Also, it will depend on if your cells can replicate the extrachromosomal DNA, like HEK-293T (with T antigen) can replicate plasmids with the SV40 ori. But normally in primary cell cultures that have not been modified to carry the supporting protein to replicate from a viral ori, the plasmid will eventually be lost.
