I am doing some experiments with the MCF7 breast cancer cells. I have established MCF7 cell lines with and without suppressing the protein of interest. Currently, I am doing cytotoxicity and clonogenic survival to Tamoxifen in complete DMEM media.
I came across quite many articles where researchers have done experiment in phenol red free media.
My question is how important it is to use phenol red free media?
How much impact would it have on the results in the context of tamoxifen sensitivity?

I have done tamoxifen studies in red media. The concentration I used was 2.8 uM and did not find any observable impact of phenol red in my investiation.

Regarding treating Tamoxifen to MCF7 cell line. Did you grow MCF 7 cells in Charcoal stripped FBS? Serum in FBS might have some impact in tamoxifen sensitivity.
I am growing MCF7 in DMEM with 10% Charcoal stripped FBS, for two passage they grow well but afterwards cells begin to die. How can we grow cells in Serum deprived condition?
