I study about isolation of environmental mycobacteria from drinking waters, but in some samples, different colonies of mycobacteria are mixed.
How I can provide pure cultures from a mixed population in mycobacteria?
I used the streaking technique, but I cannot provide pure cuitures in some samples. For example, grow of SGM after than RGM.

Your only choice to obtain pure clones is to pick single colonies from a solid medium.
I agree with Dr. Werner Solbach: Make a serial 10 fold dilution and plate for isolated colonies. Then look for distinct colonies and perform a specifies specific colony PCR thereby you are sure of the species.
First, the mixture must be diluted until the various individual microorganisms become separated that after incubation they form colonies isolated from the colonies of other microorganisms on an agar surface. Second, an isolated colony can be "picked off" and transferred to selective medium .
