Hello! I have tried to research this question all day. During a DNA isolation, we (students) used The midi kit from QIAGEN. In that protocol, they state that all centrifugation steps should be performed at 4C. Our school only had one centrifuge with cooling function available for us bachelor students (they are probably afraid we will break expensive equipment) so our group choosed to continue with regular bench top centrifuges (without cooling system) since not all groups would have time to do their centrifugation steps in that one fancy centrifuge.

I currently work on the lab report, and want to include how the differences in temperature could affect our sample for downstream application (microinjection). I assume that it prevent DNA degradation and shearing but can't find any reliable source for this. In the handbook for the kit, there is no mention of how the temperature will affect the procedure. Could anyone provide me with some clarity of how the temperature could have affected our samples?
