I am asking this because this is the only human melanoma cell line that I have had that has a blackish-brown colored pellet. Other melanoma cell lines have off-white pellet.
Thank you very much Paula and Christian for the help. I was actually looking for A375 cells transfected with luciferase for in vivo imaging. Do you have any idea about its availability?
A375 could have a very little brown pigmentation (after centrifugation) as it's melaoma (like a body spot). I used to see this phenomenom with B16/F10 (mouse melanoma cell line).
Look after the number of passages you cell culture reached, if somoene knows it (not obvious in every labs)...
To my knowledge there are no commercial A375 cells transfected with luciferase, I think you should transfect them using the luciferase kit from Promega for example.
