What is the most reliable method to measure the healing of an experimental intestinal anastomosis? The burst pressure, tensile strength, hydroxyproline concentration and histological analysis have been used but have several limitations.
Some studies use immunostaining for neovascularisation markers or collagen fibres but these reports are sporadic.

All new methods are useful for this kind of studies, However, histological analysis like morphometric parameters easy to measure & some special staining methods help you to find different parameters like collagen deposition in your healing part. i hope these brief answer help you. sincerely yours.
Thank you for your answer. I was wondering if anyone has any experience with digital morphometry and different types of immunochemistry staining for the evaluation of anastomotic healing. There aren't any reports correlating the results of these techniques with the other used techniques.
There is some data that the mechanical strength of an anastomosis as measured by the anastomotic burst pressure doesn't correlate with the hydroxyproline concentration. In a recent experimental study we performed (to be published) the anastomotic burst pressure (expressing the anastomotic strength) doesn't correlate with the histological healing parameters.

The idea is that at x days after surgery the lab animal is sacrificed and the anastomosis tested for leaking.
