Hi everyone,
Can somebody explain how I can determine the derivatization yield of analyte? I've seen lots of papers that detail their yields, but none of them mention how they calculated the derivatization yield. I read somewhere that an internal standard can also be used to calculate the derivatization yield. Hence, can someone use a practical example to explain how to calculate the yield? Or point to a really good paper that explains this?

Dear Tijana,
I've done derivatization for GC in the past, but never had to determine the derivatization yield {or %), so I can't tell you how to do this. What you might try is contacting the company that manufactures the derivatization reagent you are using and asking them your question. They should be able to tell you how to do this. The manufacturer should know this, as they are selling the reagents to GC users, so they should know how well their derivatization reagents work, and have a way of determining this. They wouldn't be in business very long if they sold derivatization reagents that didn't work well. Most chemical companies and reagent suppliers have technical service personnel to answer questions such as yours. They have to keep their customers happy so that they will keep buying their products.

From my experience, the yield of the usual derivatizations is 100%, if the criteria of time and temperature are observed. However, in the case of derivatization of multiple functional groups, it may happen that not all of them react. For example, bisphenols with 2 OH groups do not always react completely with the reagent, so that biphenol is present next to singly derivatized and doubly derivatized. In this situation, the yield can be relatively determined by the ratios of the peak areas A:B:C. Another way would be a calculation via the reagent peak. Here, the content in the sample solution can be determined via a calibration curve and thus the conversion of the reaction can be calculated.
