I use langendorff perfusion method and collagenaseII enzyme for digestion.

­­In most cases, I get high percentage of dead cells (round/oval in shape). Please advise me on what could be possible cause(s) of cell death.

And please say something about Ca2+ tolerant cardiomyocytes. What does this term mean? Why do we need Ca2+ tolerant cardiomyocytes?

And how do we condition the cells or produce Ca2+ tolerant cardiomyocytes?

I will appreciate any useful inputs or materials for reference.
