Hello: I'm planning to do ICV injection on mice (with amyloid-beta peptides for Alzheimer's disease), the dosage should be somewhere around 3µL. I'm not sure if the procedure would cause too much stress in the animals since they would be put in behavioral tests later.
How long should I let these mice recover and are there any tips on the recovery process? Thanks in advance!

Hi Lin,
The procedure is quite the same for all surgery. Be sure to have tools as sterile as you can, monitor temp, avoid any bleeding, suture and apply a local disinfectant (betadine for example), and place the mice in a warming room until wake-up. You can monitor their weight to notice any sign of infection, and I advise you wait at least two weeks before behavioral tests. Don't worry, mice usually recover well after ICV injections.

Agreed with Matthias, but make sure to inject the mice with 1µL over a timespan of 1 minute and very importantly, then wait 6 minutes before the next 1µL injection. Do this procedure until your whole volume is injected.

I fully agree with above answers but, the use of mice model for surgery is little bit difficult due to high rate of mortality. I did the same study so as per my experience i would suggest you its better to go with rat model. Because the chances of mortality will be less and recovery will be fast.

Just as a follow up.... I do not experience mortality in my ICV injections in mice. As long as you work sterile and inject the product aseptically, your lethality drops dramatically. To do this, I sterilize my hamilton syringe with 70% EtOh and leave the ethanol in the syringe until ready for injection. I rinse the syringe several times with sterile saline and then with sterile product.

I agree with Ivo, with all the procedure in a sterile environment I never experience mortality. One more detail, after the injection, you should keep the needle in place 10min to avoid any outflow of the compound and to enhance its local diffusion.
