Hello fellow researchers. I have this question regarding the preparation of 5-Aminoindole (SMAC mimetics). This is from SIGMA ALDRICH ( Catl.No.A59654 ALDRICH). I have to prepare 1mM of this.Data sheet says we have to use 0.5 % DMSO to dissolve the compound and also says its in-soluble in water. So the question is how to prepare the stock solution. Experts please help me out
I am assuming that your working concentration to test this compound is much lower than 1mM, which must be the concentrated stock, given that other SMAC mime tics are usually effective in micro molar range (if memory serves me correctly). If that is the case, simply dissolve the appropriate amount of the compound in 0.5% DMSO. I am somewhat unsure, why the question!
[Addendum: In case if you need only a small volume and do not have a microbalance to weigh sub-mg amount, prepare a 10 or 100 fold more concentrated stock and dilute appropriately for your use. At the working concentration (microM), you should be able to dilute it into the aqueous medium directly.]
