I would like to analyze whether a specific gene in my hiPS cells in methylated. Besides Bisulfidsequencing i would like to treat my cells with 5-Azacytidine, and measure expression levels before and after treatment. But i don´t know which concentration i shall use and how long i need to incubate the cells before i can detect an effect..

I use 10uM decitabine (5-aza-2'-deoxycytidine) for colon cancer cell lines and my colleagues had used the same for prostate cancer cell lines. We found this form of Aza is more stable than Azacitine. I found 50-60% of demethylation in some genes after treatment.
