Hello, I'm working on yeast two hybrid. After I mating, I usually do yeast colony pcr to amplify AD plasmid. If I test 50 yeast colonies from quadruple dropout media. I barely get 10 amplicons. I don't know how many percentage to get amplicons from yeast colony pcr. In addition, I have no idea with unamplified 40 colonies. (They grew on quadruple dropout but not amplified..) Since I have this problem, I also set up yeast colony pcr to amplify the bait construct (after mating and from quadruple drop media) . But, in that case, bait was never amplified from all yeast colonies.

Here is my pcr mixture:

Forward primer : 0.5 ul

Reverse primer : 0.5 ul

2X gotaq : 10 ul

DMSO : 0.6 ul

D.W : 8.4 ul

Total 20 ul

If you share any experience related in yeast two hybrid, that would be helpful for me.
